Lot sampling for dissolving microneedle patch arrays: what should a buyer specify?

A lot-release statement is useful only when the lot, inspection unit, defect definitions and sampling plan are clear. For cosmetic peptide microneedle patches, separate visible array defects from analytical results. A sample that passes visual inspection cannot establish peptide content or stability for every patch.
Define the lot and the unit before discussing AQL
“Check a few pieces” leaves too much open. A production lot might be defined by one formulation run, mold configuration and packaging operation, but the boundaries must be agreed in the specification. Decide whether an inspection unit is one array, one sealed pouch or one retail kit. Then define what inspectors record: incomplete tips, damaged backing, foreign matter, incorrect pouch artwork and seal appearance are different defect families. Our critical-quality-attributes guide helps identify which observations belong in a specification.
- Lot and sublot identifiers, including any packaging or mold change.
- Inspection unit and point of sampling.
- Defect descriptions supported by photographs or drawings.
- Separate disposition rules for critical, major and minor defects.
- Record of rejected units and the action taken.
Use an attribute sampling plan for the right question
ISO 2859-1:2026 describes sampling schemes indexed by acceptance quality limit for lot-by-lot inspection by attributes. AQL is a planning index for a defined inspection scheme, not a promise that every accepted lot contains no defects. The chosen sample size and acceptance number depend on the lot and plan. Buyers should ask which edition, inspection level and switching rules a supplier is using rather than accepting “AQL passed” as a complete report.
When visual sampling is the wrong substitute
Peptide content, residual moisture and dissolution are measured variables with their own methods, samples and limits. They need a different test plan. The number of visually inspected trays should not be used as proof of the peptide assay result. Likewise, a supplier may use automated imaging for broad screening but still needs to document what the camera can detect and how uncertain images are reviewed. See peptide content and uniformity testing for the analytical side.
Write a release record a buyer can audit
The FDA cosmetic GMP inspection checklist describes records for sampling, laboratory controls, results and product disposition. In an OEM agreement, identify who approves the lot, where original images and worksheets are kept, and how retests or deviations are resolved. Do not invent a universal acceptance limit for all needle counts or shapes. A crescent eye patch and a small circular array may need different defect illustrations and inspection viewpoints.
FAQ
Does AQL mean a fixed percentage of defects is permitted in every lot?
No. AQL indexes a sampling scheme over a continuing series of lots. The actual acceptance decision follows the selected scheme’s sample size and acceptance criteria.
Should every microneedle tip be inspected?
That depends on the specified attribute and inspection technology. Define what is checked on each unit, what is sampled, and how missed or ambiguous defects are handled.
Can a visual pass replace peptide assay or stability data?
No. Appearance, peptide content and stability answer different questions. Keep their sampling plans, methods and release limits separate.
