How to Protect Peptide Activity in Microneedle Patch Manufacturing
Peptide activity protection is a system: qualified raw material, compatible pH and excipients, controlled thermal and moisture exposure, suitable drying, high-barrier packaging and a stability plan. No single “low-temperature” claim is enough by itself.
Map the degradation risks
Different peptides have different sensitivities. Heat, water, oxygen, light, extreme pH and reactive ingredients can each matter. The formulation team should map plausible risks from receiving and compounding through casting, drying, demolding, packaging, transport and storage.
Control the process window
Temperature matters, but so do exposure time, water activity and oxygen. A lower temperature with a very long wet hold may not be better than a carefully controlled shorter process. Critical process parameters should connect to measurable array and formula attributes.
Make packaging part of the control strategy
Dissolvable arrays are often moisture sensitive. Pouch barrier, seal quality, headspace, desiccant strategy where appropriate, and secondary pack conditions can determine whether a good batch remains good through distribution. Package qualification should not be postponed until after the formula is fixed.
Verify instead of relying on slogans
Ask what will be measured at release and during stability. Depending on the project, this may include appearance, array integrity, moisture or water activity, dissolution behavior, microbial quality and an ingredient-specific analytical method. The test set must match the final formula and claim strategy.
